本研究的目的為探討葡萄牙牡蠣 (Crassostrea angulata) 血球在防禦機制中所扮演的角色,並建立評估細胞免疫能力的方法。化學染色結果發現,顆粒血球與非顆粒血球皆呈PAS陽性反應,而少量的顆粒血球也呈蘇丹黑B陽性反應,顯示血球的細胞質含多醣類,而部分顆粒具有脂質成分。酸性磷酸酶、鹼性磷酸酶、β-葡糖苷酸酶、α-奈酚乙酸酯酶、奈酚AS-D氯化乙酸酯酶和過氧化酶等六種酵素活性均存在顆粒血球內。評估血球免疫的反應發現,顆粒血球與非顆粒血球皆具有吞噬能力,可吞噬螢光乳珠和酵母多醣,並可產生超氧根離子來對抗外來物。血球中,顆粒血球攝取中性紅較為活躍、吞噬能力較強,其中以嗜鹼顆粒血球的吞噬螢光乳珠能力最強。

摘要abstract(英): The aims of this study were to examine the role of Portuguese oyster (Crassostrea angulata) hemocytes in the defense mechanism and to establish hemato-immunological parameters. Following cytochemical staining, the granulocytes and the agranulocytes exhibited a PAS positive reaction for polysaccharides; however, a lipid positive reaction was only observed in the granulocytes. The activities of acid phosphatase, alkaline phosphatase, β-glucuronidase, naphthol AS-D chloroacetate esterase, α-Naphthyl acetate esterase and peroxidase were observed in the granulocytes. In immunological assays, both of the granulocytes and the agranulocytes were able to phagocytose fluorescent latex beads and zymosan particles and produce superoxide anion against foreign agents. Moreover, the granulocytes exhibited greater phagocytic ability and more activity in the uptake of neutral red than agranulocytes. The basophilic granulocytes showed the strongest phagocytotic activity of fluorescent latex beads.
