臺灣北部海域鎖管漁業資源之調查評估

劍尖槍鎖管 (swordtip spear-squid, Uroteuthis edulis) 為臺灣燈火漁業的重要漁獲物種之一。由於頭足類資源多受環境影響而呈現年間波動,因此易受過度捕撈的影響,導致資源的崩潰,凸顯資源評估之重要。本研究以環境DNA作為指標,開發一種新流程,以從水量較小且未經過濾的海水樣品中提取eDNA。並將其應用在臺灣北部海域鎖管漁獲資料的校正。使用修改後的程序,萃取eDNA所需的樣本水量減少到25ml,同時無需經過過濾,只需保存在-20°C即可。基此,研究人員採取水樣將更為便捷,同時亦提高漁民和研究人員合作收集水樣的意願。此一改良的程序無須過濾,同時可以降低eDNA的損失,並且避免了過濾水樣,濾材阻塞的影響。另一方面,樣本分析結果顯示,主要鎖管漁獲物種中,劍尖槍鎖管出現在東海陸棚區,而杜氏鎖管則主要出現在中國沿海,相對來說,中國槍鎖管eDNA出現比例較低。

Uroteuthis edulis is one of the important catch species in Taiwan troch light fishery. Because cephalopod resources are mostly affected by the environment and show annual fluctuations, they are susceptible to overfishing, leading to the collapse of resources and highlighting the importance of resource assessment. This study uses environmental DNA (eDNA) as an indicator to develop a new process to extract eDNA from a small amount of water and unfiltered seawater samples. And it is applied to the correction of troch light fishery catch data in the northern waters of Taiwan. Using the modified procedure, the amount of sample water required to extract eDNA was reduced to 25ml, and no filtering was required, and it only needed to be stored at -20 °C. Based on this, it will be more convenient for researchers to take water samples, while also increasing the willingness of fishermen to collect water samples for researchers. This improved procedure does not require filtration, at the same time reduces the loss of eDNA, and avoids the effects of filtering water samples and blocking the filter material. On the other hand, the sample analysis results show that among the main troch light fishery species, U. edulis, appear in the continental shelf area of the East China Sea, while Uroteuthis duvauceli appear mainly along the coast of China. The percentage of eDNA in Uroteuthis chinensis is low.
