全雌化烏魚種苗繁殖技術之研發

烏魚養殖是台灣重要漁業之一,由於烏魚子價格高,養殖業者便以添加雌激性素使 烏魚雌性比例提高到90%以上,而雌性激素的大量使用引發生態及食品安全的疑慮 ,故需朝建立全雌化養殖烏魚技術發展,以達減少使用雌性激素促進烏魚變性之機 會。本計畫主要將進行培育雄性化烏魚產精,建立烏魚精液凍結保存,在烏魚繁殖 季節進行人工繁殖並培育子代,由子代性別比例判別親代是否為新雄魚。新雄魚在 與一般母魚配對繁殖,生產出全雌化烏魚種苗。本研究結果,產精僅有6尾,平均產 精量約1ml,可供人工繁殖及冷凍保存用。新雄魚會逆轉回雌魚,且有精、卵巢並存 的中間性別階段(intersex)。有7尾雄性化烏魚(已標示晶片)之子代(已1年齡)(圖 3),需培育養殖至第2繁殖季(2年齡),才能作為判別親魚是否為新雄魚之依據。另 外,有4尾雄性化烏魚在今年逆轉回雌魚,其子代的性別為何? 將能驗證本研究的最 初推論-『以新雄烏魚繁殖生產全雌化烏魚魚苗』 進行人工受精,繁殖孵化及培育 魚苗。雌烏魚誘導催熟、擠卵、受精、孵育及受精卵與胚胎發育過程,此次僅培育 出3尾雄性化烏魚子代。

Grey mullet culture is one of the important fishery in Taiwan. In order to produce expensive mullet roe, fisherman feed fish juvenile with estrogen-containing feed to improve female ratio more than 90%. Estrogen is a chemical compound that is a ecology pollutant and cause the doubt of food safy of mullet roe. We need to develop new technique for production of all female grey mullet progeny to avoid using estrogen. We have already masculized grey mullet and begin spermiation in the present. The following steps in this study are: 1. semen cryopreservation for artificial propagation and culture the progeny. 2. check the sex of offspring to confirm the XX-type male breeder(neomale). Finally, we can use neomale breeder to propagate all female gret mullet progeny.In this study, 3 masculized grey mullet begin spermiation and semen cryopreservation have finished. We also induce female gret mullet spawning, eggs fertilized with masculized grey mullet semen and hatching and larvae culture.
