紅葡萄藻(Botryocladia leptopoda)生物活性成分之應用研究

海藻因含有豐富營養素及各式礦物質,且具有多項的生理活性功能,因而有「長壽 菜」的美稱,據文獻指出紅葡萄藻(Botryocladia leptopoda)具有強抗氧化能力 ,因此本研究預萃取其多酚,以開發傷口敷料進行商品評估。紅葡萄藻( Botryocladia leptopoda) 利用EtOH 60;EtOH Freeze dry;Alkaline-EtOH;EtOH 70;H2O Sterilization不同的製程萃取剩餘藻渣與原藻中的多酚化合物。其中以冷 凍乾燥紅葡萄藻進行萃取的製程中保有較高的多酚,鹼萃紅葡萄藻剩餘藻渣的萃取 製程則次之。三種抗氧化指標,還原力、DPPH清除自由基、螯合亞鐵離子,只有還 原力不表現。抗發炎活性評估中,IL-6表現變化量則為EtOH Freeze dry(-2189%) 與Alkaline-EtOH(-1673%),顯示EtOH Freeze dry及Alkaline-EtOH對已發炎細胞具 有調控作用。轉化生長因子TGF-1β在EtOH Freeze dry(45.40%) 與AlkalineEtOH(51.43%)皆有提升,顯示萃取物具有組織修復的潛力。而利用L929小鼠纖維母 細胞加入紅葡萄藻Alkaline-EtOH和EtOH Freeze dry產物經H2O2誘導後進行抗氧化 SOD、Glutathione、Catalase Activity試驗,只有在SOD試驗中 EtOH Freeze dry產物有顯著還原超氧化物(superoxide,O2-),移除過多自由基的可能性。綜 觀以上,紅葡萄藻在各項抗氧化、抗發炎試驗,可作為後續發展的評估。

Seaweed is often regarded as a “longevity delicacy”, because it is enriched in critical nutrients and minerals, which is thought to be beneficial to humans in physiological terms.  It has been reported that extract from Botryocladia leptopoda, a kind of red alga, has strong antioxidant activity.  We therefore propose here to extensively characterize the polyphenols containing in B. leptopoda for its potential activity in wound healing. We extract the polyphenol compounds from B. leptopoda and its residue in different procedures : EtOH-60; EtOH-Freezedry; Alkaline-EtOH; EtOH-70; and H2O sterilization. And the extraction product from EtOH-Freeze-dry contains much more polyphenols, the extraction product from Alkaline-EtOH taked a seconed place. Polyphenols from B. leptopoda can work as anti-oxidation in the 3 kind of index: reducing power, DPPH radical scavenging ability and ferrous ion chelating ability, but it is less effective in reducing power. In anti-inflammatory activity evaluation, the IL-6 variations in EtOH-Freeze- dry and Alkaline-EtOH are -2189% and -1673%, showing that these products play as a regulatory role in inflammatory cells. In the other ways, the concentration of transforming growth factor-1beta (TGF-1β) rise in both EtOH-Freeze-dry (45.40%) and Alkaline-EtOH (51.43%) groups, means the extracts has the potential for tissue repair. Add B. leptopoda extractions from EtOH-Freeze-dry and Alkaline-EtOH in the culture medium of mouse fibroblast cells L929, before add H2O2 .We can observe the concentration of execute superoxide dismutase (SODs) only increase when EtOH-Freeze-dry product exists. Obviously it has possibility of  redox reducing superoxide(O2 - ), which can remove excessive free radical. Our long-term goal is to promote and maximize the utilities and the economic values of algae, thereby furthering the establishment of a vibrant algal industry in Taiwan.
