褐藻作為改善肌肉流失之營養膳食補充品研發(2)

銅藻(Sargassum horneri)、冬青葉馬尾藻(Sargassum ilicifolium)與中國半葉馬 尾藻(Sargassum hemiphyllum var. chinense)萃取物250μg/mL添加於小鼠骨骼肌 細胞C2C12中可以使其分裂量上升為1.42倍、1.74倍與1.77倍。分析其中促肌肉生長 因子4EBP1磷酸化程度分別為控制組的1.12倍、1.22倍與1.20倍;S6K1磷酸化程度則 為控制組的0.98倍、1.19倍與1.18倍,足見萃出物有效刺激肌肉生長。對此項肌肉 生長路徑之上游調控因子Rag A/B進行西方墨點法分析,並將蛋白質表現量化,顯示 加入褐藻萃出物後較控制組上升約70%,確認褐藻產物能透過開啟肌肉生長路徑來增 加肌肉表現。另以慢性病常用藥物Dexamethasone來誘發肌肉細胞萎縮,測試骨骼肌 細胞C2C12中的肌肉降解因子Cathepsin-L、myostatin、MuRF1等,發現各種馬尾藻 產物至少對1項指標有所改善,表示亦可透過降低藥物副作用的方式來達到肌肉保護的目的。

The addition of 250 μg/mL of Sargassum horneri, Sargassum ilicifolium and Sargassum hemiphyllum var. chinense extract to mouse skeletal muscle cells C2C12 can increase the number of cells to 1.42 times. , 1.74 times and 1.77 times.
