文蛤遺傳多樣性之調查研究-建構生命條碼資料庫

文蛤為臺灣重要經濟水產貝類,屬於軟體動物門雙殼綱簾蛤科文蛤屬。但進行物種 分類時很難只依其外殼形態判別,必須搭配DNA序列資料。本計畫主要是調查臺灣文 蛤物種多樣性,並建立文蛤之生命條碼,闡明種屬間的親緣演化關係,協助文蛤的 繁養殖、保種、育種等相關研究及管理。 以coI基因為目標所設計引子擴增DNA片段 為631bp,蒐集來自新北市、彰化縣、雲林縣、嘉義縣、臺南市、高雄市及金門縣養 殖及野生文蛤樣品,共計完成161筆生命條碼之PCR及定序分析。本研究也首次發現 已coII基因序列為目標片段,可快速的區分M. lusoria與其他4種文蛤,透過粒線體 coI及coII基因序列的比對分析及親緣性分析,臺灣目前主要的文蛤為M. lusoria。

Hard clams is one of the important farmed shellfish in Taiwan. It belongs to the phylum Mollusca, class Bivalvia, order Veneroida, family Veneridae. Only depending on the shell morphology is difficult to identify them correctly. However, much of the clam classification to date is based on shell characteristics. In order to identify them correctly, we have to work with DNA sequence data. The purpose of this project is to investigate the diversity of the hard clams in Taiwan, constructing the database of bio-barcode, clarifying the phylogenetic relationship and to assist the breeding study and conservation management. The bio-barcode fragment is 631bp, the PCR primer was designed form the coI gene sequence. The hard clem samples were corrected from the New Taipei City, Changhua, Yunlin, Chiayi, Tainan, Kaohsiung and Kinmen. Total 161 biobarcode sequence were corrected in the bio-barcode database.  In this study, we first report that the M. lusoria could be identified quickly by coII gene sequence. Through the sequence aligment and phylogenetic analysis of the mitochondrial coI and coII gene sequences, M. lusoria is the main current hard clam species in Taiwan.
