水產種苗研究團隊-雞卵黃抗體應用於龍膽石斑種苗培育之研究

本研究以評估抗石斑虹彩病毒之卵黃免疫球蛋白被動保護效果。以石斑魚泳鰾細胞大量增殖石斑虹彩病毒 (grouper iridovirus of Taiwan, TGIV),再以PEG (Polyethylene glycol) 濃縮並進行蔗糖不連續梯度之病毒純化,將純化的台灣石斑虹彩病毒以肌肉注射免疫雞隻,並取蛋黃液以酵素連結免疫吸附分析(ELISA)IgY抗體力價,結果在免後三個月才開始正常產蛋,蛋液抗TGIV之IgY力價在102~3之間。

In this research, yolk of immunoglobulin against TGIV was prepared and its effectiveness on passive protection. Massive multiplication grouper iridovirus of Taiwan (TGIV ) by the grouper swimming bladder cell cultured.In order to get adequate TGIV antigen ,the concentration used PEG (Polyethylene glycol) and purification from the sucrose discontinuous gradient ultracentrifugation. The chicken immunized by intramuscular injection with purified TGIV, and to analyze the IgY antibody titer by ELISA method, the result in laying egg start at three months after immunization .The titer of IgY in yolk were between 102 to 103.
