魚類神經壞死症病毒(Nervous necrosis virus, NNV)是造成許多海水養殖魚類的重要病原,包括石斑魚在內,又早期魚苗是主要感染及死亡的時期。根據已發表之文獻,可感染神經壞死症之魚種皆有垂直感染NNV病毒的案例存在,目前已發表的防治方法包括洗卵及淘汰帶原種魚。本計畫目標則在評估NNV去活化疫苗對誘發石斑種魚產生中和抗體的效果,期由免疫石斑種魚來達到大幅降低或阻斷種魚體內病毒垂直傳染魚卵的機率。第一年試驗內容,乃使用點帶石斑成魚(平均體重1.65 kg)進行最佳疫苗免疫劑量與條件的測試。以兩種劑量的NNV死毒疫苗免疫石斑成魚後,每兩週採血一次,測中和力價,已連續追蹤五個月;結果顯示,以109 TCID50 kg-1之免疫劑量比108 TCID50 kg-1 引起之中和抗體力價高,且使用佐劑組比無佐劑組效果好,以肌肉注射比腹腔注射組安全。
本年度對長期篩檢培育之鞍帶石斑種魚進行3次催熟產卵生產試驗,共獲卵重23.33Kg,其中胚體卵2.22Kg。並進行3 批次之瑪拉巴石斑、七星斑及點帶石斑育苗生產試驗,育苗率分別為3.1%、8.01%及28%,共生產石斑類白身苗132250尾。
淡水中心研發成功的新雜交品系,只要1年即可達上市體型,且雄性比率高,是極適合做為推廣養殖的優質品系。計畫預計生產魚苗10萬尾提供業者試養,本年度已生產魚苗約23萬尾配售給3戶業者試養,但因莫拉克颱風造成魚苗流失。另已培育尼羅與歐利亞魚苗各一批,作為明年度挑選種魚,以加入魚苗大量生產。
以6條引子(p3、p5、p7、p8、p9、p12)檢測90個不同來源的養殖九孔族群,共有81條帶,利用UPGMA群集分析,結果當遺傳距離0.3,可將90個樣本劃分為4群。另從100個逢機引子篩選出產物清楚且重覆性高的引子6條 (p24、p31、p36、p39、p40、p48),檢測60個紅色吳郭魚hcp品系,TsR品系與引進種cp雜交後代,結果平均p24有4條擴增帶,p31有2條,p36有6條,p39有3條, p40有3條,p48有7條,總共有25條擴增帶,利用UPGMA群集分析,結果當遺傳距離0.22,可區別紅色吳郭魚TsR品系與引進種(cp及hcp)品系及大部分的ct品系。

The aim of this plan is evaluating NNV inactivated vaccine to induce the broodstock to produce the neutralizing antibody. We wish to reduce or block the roe of NNV vertical infection. The first year, the adult fish (average body weight 1.65 kg) was used to carry on the best vaccine immunizing dose and the condition. After immunity by two dosage's NNV inactivated vaccine ,To measure and trace the titer of neutralized antibody continuously for five months. The result showed that the titer is higher in dose of 109 TCID50 kg-1 than 108 TCID50 kg-1, and adjuvant group is better than not adjuvant group , the security of injection by intramuscular compared to abdominal cavity injection group.
The newly hybrid tilapia, bred by Freshwater Aquaculture Research Center, FRI, only need to be cultured for one year to marketable size, with high male ratio, are very suitable for the farmers.Previously, 0.1 million fry were to be produced this year, but total of 0.23 million had been already sold to 3 farmers; unfortunately, the cultured fry were all run off by the heavy rainfall from Typhoon Morakot, Fry of Nile tilapia (Oreochromis niloticus) and Blue tilapia (Oreochromis aurea), respectively, were cultured to be selected as broodstock next year.
Cultured small abalone, Haliotis diversicolor, collected from six hybrid combinations the sum of 90. RAPD and six random primers (p3、p5、p7、p8、p9、p12) were then used to analysis and amplify 81 clear and reproducible bands, then utilize Un-weighted Pair Group Method Analysis (UPGMA), when the genetic distance 0.3 can divide 90 samples into 4 groups.
Red tilapia, Oreochromis spp, collected from one hybrid (ct), inbred lines (TsR) and commercial line (cp and hcp) the sum of 60. RAPD and six random primers (p24、p31、p36、p39、p40、p48) were then used to analysis and amplify 25 clear and reproducible bands, then utilize Un-weighted Pair Group Method Analysis (UPGMA), when the genetic distance 0.22 can divide TsR line and commercial line: cp, hcp, a lot of ct.
