The purposes of chromosome manipulation R&D are to establish the species specificoperational protocols of chromosome manipulation, to promote the biotechnology ofchromosome manipulation, and to provide the private sector with mature and wellestablishedtechnology in this aspect. Chromosome manipulation protocols will befurther studied to ensure the ideal combination of satisfactory hatching rate and triploidpercentage. For early determination of the percentage of triploids in shellfishes , earlylarvaein addition to hemocyte samples from the adductor muscle in late larvae or adultwill be further tested with the purpose to decide whether the population is with desirabletriploid percentage. The procedure included (1) staining the cell suspension with afluorescent dye such as Hoechest 33342 or propidium iodide that specifically bindsDNA and RNA, (2) removal of RNA by application of RNase, and (3) determining thefluorescence upon mercury excitation in Partec PA flowcytometer. The optimal lowlevel, flow rate, laser power and bitmap gating or gain value in flow cytometer will beadjusted to meet the required conditions for various samples.An automatic manipulation system for the mass propagation and chromosomemanipulation of the small abalone, Haliotis diversicolor, Pacific oyster, Crassostrea gigasand other economically important shellfishes will be studied in terms of improvedfunction, better parts, more flexible to meet the requirement of various species. Thesystem will be further designed based on the previous try and error of optimumconditions obtained from tests simulating the possible conditions during automaticmanipulation and the species-specific characteristics of unfertilized and fertilized eggs invarious experimental shellfishes. The improvement of the important control box is alsoemphasized with the purpose to omit any possible mistake.due to the operatingindividuals or unique coastal environment. The new design of the automaticmanipulation system will incorporate all the necessary features. The automatic controlbox of the system therefore will ensure various specific treatments, such as those for mass propagation and chromosome manipulation. As a whole, the system will be moreapplicable for regular mass propagation and chromosome manipulation procedures ofthe shellfish than the first one of its kind.Key words:shellfish, mass propagation, chromosome manipulation, automation.
