鱟血清成份應用於熱原檢測試劑之開發

美洲鱟血液的變形細胞可與細菌內毒素結合產生凝集,因此早已被開發為測定內毒素的國際標準方法。我國目前雖在金門及澎湖沿海擁有野生中國鱟之自然資源,但尚未開發成商業化生物製劑。隨著海洋資源的耗減,野生鱟的產量日益降低,未來養殖鱟將會成為趨勢。然而鱟的養殖需要10~15年才能成年,故增加養殖的難度。再加上鱟的血液成份有明顯的季節差異,隨著季節的變化,血液中的成分會有明顯的差異,使養殖鱟無法一年四季採血,並影響其生物製劑的靈敏性及產量。本計畫擬檢測飼養的三棘鱟血液中變形細胞與季節變異的關聯性,以做為未來生產製造鱟檢測試劑時的採血量依據。更進一步評估水溫等環境因子,是否使其血液成分有所變化。鱟試劑成功的開發,將可帶來國際商機。

Limulus amoebocyte lysate (LAL) clots when mixed with endotoxins which are lipopolysaccharide components of the outer cell-wall layer of gramnegative bacteria. Since the initial description of the lysate method for the detection of endotoxins, it has become an international standard method. Although Jinmen and the Penghu coast has the wild Chinese horseshoe crab, but not yet developed into the commercialization of biological agents. With the depletion of marine resources, wild king crab production declining, the future cultured crab would become the tendency. However, the culture of Chinese horseshoe crab (Tachypleus tridentatus) needs 10 ~ 15 years to adult, it would increase the difficulty of culture. In addition, the blood components have obvious seasonal variation, with seasonal changes in the composition of the blood there will be significant difference. So it would not to collect Limulus blood throughout the year and affect its biological agents and yield sensitivity.
