葡萄藻機能成分萃取及其應用

本試驗利用葡萄藻酵素水解萃取物進行血管收縮素轉換酶(ACE)抑制試驗,葡萄藻酵素水解萃取物ACE抑制活性試驗,結果顯示抑制效果Pepsin > Flavourzyme > Alcalase,尤其pepsin水解萃取物ACE抑制效果顯著高於其他2個酵素萃取。葡萄藻酵素水解萃取物MTS試驗中以RAW264.7之老鼠巨噬細胞試驗毒性分析,試驗結果顯示對巨噬細胞無毒性產生。葡萄藻葡萄藻酵素水解萃取物體外抗腫瘤細胞株試驗,結果顯示葡萄藻酵素水解萃取物對於小鼠血癌細胞WEHI-3及人類血癌細胞株HL-60,皆具有良好抑制效果,且由彗星試驗觀察DNA拖尾程度,證明葡萄藻酵素水解萃取物對血癌細胞具有損傷的效果。

In our study, the ACE inhibitory activity of Caulerpa microphysa enzyme-hydrolysed were assay, the results shown that the inhibitory effect was in order: Pepsin> Flavourzyme> Alcalase, especially pepsin extraction. The pepsin extraction was significantly better than the other enzyme extraction. The MTS((3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium)) toxicity tests of Caulerpa microphysa enzyme extracts were tested by RAW 264.7 cell line. The results showed there was no toxicity to macrophage. For in vitro anti-tumor test, enzyme extracts of Caulerpa microphysa were assay for mice WEHI-3 cell line and human HL-60 cell line. The results shown which have well inhibitory effect and Caulerpa microphysa enzyme extracts was induces the DNA damage in WEHI-3 cell by comet assay.
