水產種苗研究團隊-龍膽石斑遺傳育種之研究

本研究目的主要為建構選種及育種模式建立,加強龍膽石斑魚成長育種,本年度執行目標為優良種魚持續選拔,試驗結果顯示目前經選拔後魚隻(第A1組)平均體重為14.2kg±0.34體長為82±1.51cm,經淘汰後魚隻平均體重為8.4kg±0.54體長為80±1.23cm,在型態上具有明顯差異;在建立龍膽石斑功能性基因選殖方面,成功選殖MEF2D(Myocyte enhance factor 2)、AKT2(Protein kinaseB)與蛋白質合成及肌肉成長相關基因,並進行在組織分佈相對定量分析,結果顯示MEF2D在尾鰭、鰓、肌肉、脾臟及鱗片中都有較高的表現,另外AKT2也有類似的結果,最後在遺傳特性資料建立上面,初步建立多對已進行篩選過的微衛星SSR引子,而(A4組)該組具成長優勢基因型魚隻將持續蓄養保種,該篩選過的微衛星引子可供後續多批次進行分析。

The main purpose of this study, to construct the mode of selection and breeding to establish, strengthen Gentian grouper grow breeding operation platform, this year's performance goals for the continued excellent species of fish selection. The test results show that after selection, the fish (A1) average weight of 14.5kg body length 82cm, after elimination fish average weight of 10.4kg body length of 80 cm, with a significant difference in the patterns. Establish the giant grouper functional gene cloning, the successful cloning MEF2D of AKT2 TGF beta receptor and protein synthesis and muscle growth related genes, and the relative quantification of tissue distribution. The results showed that MEF2D has higher performance in the caudal fin, gill, muscle, spleen, and scales. AKT2, TGF beta receptor also had similar results, and finally in the genetic characteristics, above the initial establishment of the multi on microsatellite SSR primers screened, the batch with the growth advantage genotype fish only will last Keeper conservation, the microsatellite primers available for follow-up over the multi-batch analysis.
