快速鑑別鱸鰻與短鰭鰻技術之研發―環式恆溫擴增法之應用

本研究應用環式恆溫擴增法 (loop-mediated isothermal amplification, LAMP) 鑑別台灣地區之鱸鰻 (Anguilla marmorata) 與短鰭鰻 (A. bicolor pacifica)。利用粒線體16S rRNA片段,設計對鱸鰻具特異性之LAMP引子作為工具。為了解此LAMP方法之敏感度,以不同含量之細胞全DNA測試,結果顯示以LAMP方法恆溫擴增,僅 1 pg DNA就能在短時間成功的利用電泳或可見螢光偵測到。因此,利用LAMP方法,可有效且快速的區別出鱸鰻與短鰭鰻。實驗顯示LAMP方法能在短時間 (約1小時) 鑑定鱸鰻,較一般PCR鑑別方法快約1 ~ 2倍的時間,且LAMP具有高靈敏性,可偵測極微量之DNA,此技術可能在其他魚類鑑定上有更多的應用空間。

Loop-mediated isothermal amplification (LAMP) method was employed to identify Anguilla marmorata and A. bicolor pacifica. Partial sequences of mitochondrial DNA 16S rRNA were used to design LAMP primers that were specific to A. marmorata. To test the sensitivity of the LAMP method, different amounts of template DNA were used. It showed that 1 pg of the total template DNA could be detected by both electrophoresis and visual florescence. The result showed that A. marmorata could be easily distinguished from A. bicolor pacifica within an hour. This suggested that LAMP assay could be used to identify other species.
