海洋乳酸菌之分離與鑑定

本研究採集台灣東部知本外海不同水深之海水 (285 m - 912 m)、黑鯛 (Acanthopagrus schlegeli) 腸道及本所海水吳郭魚 (Oreochromis niloticus) 養殖池水樣品,進行海洋乳酸菌之篩選、分離與鑑定。結果顯示,自深層海水測站ST14 水深334 m 及525 m,各分離與鑑定出1株乳酸菌,編號分別為ST14-334 LA3 及ST14-525 LA1,分離自黑鯛腸道之2株乳酸菌,編號分別為SB1及SB8,而本所海水吳郭魚養殖池水樣品所分離之1株乳酸菌,編號為B3。依據菌株的各項生理、生化特性、對於各種碳水化合物之利用性,以及16S rRNA 基因序列測定以鑑定該5株乳酸菌,分別將ST14-334 LA3 及ST14-525 LA1菌株歸屬為Enterococcus faecium 及 Enterococcus gallinarum;將SB1及SB8菌株歸屬為Weissella cibaria 與 Leuconostoc citreum;B3菌株歸屬為Leuconostoc pseudomesenteroides。
研究報告摘要(英) Isolation and identification of marine lactic acid bacteria (LAB) from deep ocean water, intestine of black sea bream (Acanthopagrus schlegeli) and sea water reared tank of tilapia (Oreochromis niloticus) were conducted. Eight samples (from 285 m to 912 m) of deep ocean water were collected from Chinpen (eastern Taiwan) off-shore. Among the 136 isolates from the deep sea water, 2 isolates were identified as LAB. The 2 isolates, named ST14-334 LA3 and ST14-525 LA1, were sampled from the depth of 334 m and 525 m of the station 14, respectively. Two isolates from the intestine of the back sea bream were identified as LAB. The 2 isolates, named SB1 and SB8. The strain named B3, was isolated from the sea water reared tank of tilapia. Based on the physiological, biochemical characteristics and 16S rRNA sequence analysis, the ST14-334 LA3 and ST14-525 LA1 were identified as Enterococcus faecium and Enterococcus gallinarum, respectively. The SB1 and SB8 were identified as Weissella cibaria and Leuconostoc citreum, respectively. The B3 was identified as Leuconostoc pseudomesenteroides.
