生物技術在九孔及吳郭魚之應用研究

本計畫分成兩部分,(1)吳郭魚(Oreochromis sp.);(2)九孔(Haliotis diversicolor)。吳郭魚已建立紅色吳郭魚品系(TsR),並比較F1、F2及F3三個世代的三月齡魚的體長及體重,其體長及體重的平均值隨著世代數的增加而微幅的下降,體長方面在世代間無統計上的顯著差異(p>0.05),體重方面F1及F2沒有顯著差異(p>0.05),但F3與F1及F2則有顯著差異(p<0.05)。從F2中選拔體長最大及體長最小者各自配對結果,體長的選拔是有明顯效果,同時亦以RAPD的檢測模式比較體長最大和體長最小的仔代其基因型在分子量約500bps位置的差異;體色在分子量約450bps位置的差異。九孔比較台南群自交與台南和平島雜交一月齡及六月齡活存率,台南群自交F1及F2兩世代之浪板上一月齡活存率分別50.25±6.64%及41.45±6.69%,經waller-Duncan K-ratio t test檢測並沒有顯著差異(p>0.05),六月齡活存率分別0.42±0.15%及0.15±0.07%,F1顯著優於F2(p<0.05);雜交貝(台南/和平島) 在一月齡及六月齡活存率(72.70±3.72%,0.83±0.14%)經waller-Duncan K-ratio t test檢測均顯著優於台南自交的F1及F2(p<0.05)。同時以野生九孔進行微隨體之基因選殖,至目前共10組,其中重覆主體(CA)n型態5組,(GA)n 2組,(TC)n、(GT)n 及(TGAG)n 各一組。關鍵詞:吳郭魚(Oreochromis sp.);九孔(Haliotis diversicolor);Waller-Duncan K-ratio t test;微隨體;重覆主體;基因座
研究報告摘要(英) In this study, there were two parts, (1) Tilapia (Oreochromis sp); (2) Small abalone (Haliotis diversicolor).Tilapia:We successfully established one progenies and pedigree of Tilapia, named TsR. Comparison the body length and weight of three-months-old between F1, F2, and F3 three generations, the mean of body length and weight was slightly declined with increasing number of generation. However, in statistics there was no significant difference (p>0.05) in body length nor in the body weight between F1 and F2, but there was a significant difference (p<0.05) between F2 and F3. In F2 generation, according to the body length was divided into three groups the biggest-sized, medium-sized and smallest-sized, chosen as parents from the biggest-sized and smallest-sized group, respectively. Offspring of groups was a significant difference (p<0.05), utilized specific gene marker compare the biggest-sized and smallest-sized group, there was a significant association with body color and growth rate at molecular weight 450 bps and 500bps, respectively. Small abalone:Comparison the Tainan and hybrid (Hopin(♀) cross Tainan(♂)) group of survival rate at one-month-old and six-month-old. The one-month-old and six-month-old survival rate of F1 and F2 in Tainan were 50.25±6.64% ; 41.45±6.69% and 0.42±0.15% ; 0.15±0.07%, respectively. Analyzed by waller-Duncan K-ratio t test, there was not a significant difference (p> 0.05) at one-month-old, but a significant difference (p< 0.05) at six-month-old, this result showed that F1 was superior to F2 (p<0.05). The survival rate of one-month-old and six-month-old in hybrid group were 72.70±3.72% and 0.83±0.14%, comparison the Tainan and hybrid group, analyzed by waller-Duncan K-ratio t test, there was a significant difference (p> 0.05) between the Tainan and hybrid group. Besides, we cloned microsatellite from wild small abalone of the sea area along the north of Taiwan. So far, 10 loci were cloned and sequence, there were 5 loci of (CA)n repeat motif, 2 loci of (GA)n, the rest (TC)n, (GT)n and (TGAG)n were one respectively.
