水產種苗研究團隊-台灣紅色吳郭魚之育種研究

選取去年度第一次回交子代中之紅色帶黑點個體實施自交 (A) 或與純紅吳郭魚實施第二次回交 (B),另以純紅吳郭魚之第三次自交作為對照組 (C),比較各組子代之體色與成長情形。應用微衛星DNA生物技術分析子代之遺傳變異情形。
結果顯示各組皆有純紅吳郭魚出現,A組有22尾 (純紅比率7.3%) ,B組23尾 (7.7%),C組28尾 (28%),其平均體長、平均全長與平均體重,都是A組顯著地小於B組及C組 (p < 0.05),於B組及C組之間則無顯著差異 (p > 0.05)。
分析UNH123、UNH216、UNH222、UNH846、UNH849及UNH860等6個微衛星DNA基因座之變異,結果在觀測異型合子比例 (Ho) 及期望異型合子比例 (He) 之平均值方面,C組子代分別為0.43及0.38,均小於A組子代 (0.61, 0.48) 及B組子代 (0.61, 0.53),顯示其遺傳變異性較低。
研究報告摘要(英) Red-with-black-spots tilapia of the first backcross progeny were selected to implement self cross (group A) or second backcross with pure-red Taiwanese red tilapia (group B), and the third self cross of pure-red Taiwanese red tilapia (group C) was as a control. For the progeny from each group, growth and body coloration were compared, and genetic variations were analyzed with microsatellite DNA. Results showed that pure red tilapia were bred in all three groups, number and ratio of pure red tilapia were 22 (7.3%) for group A, 23 (7.7%) for group B, and 28 (28%) for group C, respectively, and the mean body length, total length and body weight of group A were all significantly less than those of group B and group C (p < 0.05), while the last two groups were not significantly different with each other (p > 0.05). Microsatellite DNA analysis of UNH123, UNH216, UNH222, UNH846, UNH849 and UNH860 locus showed that both mean observed heterozygosity (Ho) and mean expected heterozygosity (He) for progeny of the control were all smaller than those of the experimental groups, the former was with less genetic variation.
