水產種苗研究團隊-台灣重要養殖魚貝類之寄生蟲生態防 治研究

本計畫將自點帶石斑收集不同生活史時期的卵圓鞭毛蟲,以無菌操作的方式分離、
純化各蟲體並維持繼代。10種以上不同的抗凍劑將進行對不同生活史蟲體的毒性試
驗,繼而嘗試找出無毒抗凍劑的最佳抗凍濃度。最後,藉由觀察營養體的活存率、
孢囊體的分裂情形及浮游幼體的運動性來比較出各蟲體最適的低溫保存條件。本年
度計畫的目標即為建立卵圓鞭毛蟲保種及繁殖方法,讓後續研究計畫所需的試驗材
料不虞匱乏。
研究報告摘要(英) The Amyloodinium ocellatum was collected from grouper (Epinephelus coioides). Different stages of the parasite were isolated and maintained. The aseptic manipulation and cultivation of tomonts, trophonts and dinospores were conducted. This project will firstly assay the toxicities of about 10 different cryprotectants. The optimal concentration of the cryprotectant without toxity to the parasite will secondly be considered. Finally we will test different cryopreservations including 4℃ refrigeration, -20 ℃freezing , programming freezing (-1 ℃ min-1)and directly chilling in liquid nitrogenfor different stages of the parasites with different stages of Amyloodinium ocellatum. Survival of trophont, morphology and division of tomont and motility of dinospore were evaluated to compare the effects of cryopreservations mentioned above. The aim of this study is to set up the preservation and reproduction methods to Amyloodinium ocellatum for the continuously supplying the experimental materials for the following projects.
