褐藻作為改善肌肉流失之營養膳食補充品研發(1)

   銅藻(Sargassum horneri )、冬青葉馬尾藻(Sargassum ilicifolium )與中國半 葉馬尾藻(Sargassum hemiphyllum var. chinense)進行水解條件探討,結果顯示以 1%粉碎乾燥藻體以121℃高壓熱萃30 min後,添加醣解酵素在50 ℃下反應180 min為 較佳水解條件,此時銅藻(S. horneri )之leucine、isoleucine、valine分別為 0.85mg/g、0.15mg/g、0.35mg/g;冬青葉馬尾藻(S. ilicifolium )之leucine、 isoleucine、valine分別為0.90mg/g、0.24mg/g、0.72mg/g;中國半葉馬尾藻(S. hemiphyllum var. chinense)之leucine、isoleucine、valine分別為0.65 mg/g、 0.46mg/g、1.01mg/g,三者皆具有支鏈胺基酸。在細胞存活率方面,褐藻萃取物對 老鼠骨骼肌細胞(C2C12)之存活率以銅藻(5mg/mL)大於中國半葉馬尾藻(5mg/mL)大於 冬青葉馬尾藻(10mg/mL),在銅藻、中國半葉馬尾藻、冬青葉馬尾藻各劑量濃度5 mg/mL時細胞存活率分別為82.24%、97.22%與136.60%。在細胞活性分析部分,三種 褐藻(銅藻、中國半葉馬尾藻、冬青葉馬尾藻)之mTOR表現量,分別為97.20pg/mL、 21.72pg/mL與100.52pg/mL相較於未添加組具有增加的趨勢且具有劑量依賴性。

 The hydrolysis process of Sargassum horneri ,Sargassum ilicifolium , and Sargassum hemiphyllum var. chinense was discussed, and the results showed that 1% crushing dry algae extract with 121℃ of 30min, glycolytic enzyme, 50℃ of 180min as the best hydrolysis process, at which time the leucine, isoleucine and valine of S. horneri were 0.85mg/g, 0.15mg/g, 0.35mg/g; thee leucine, isoleucine and valine of S. ilicifolium were 0.90mg/g, 0.24mg/g, 0.72mg/g, the leucine, isoleucine and valine of S. hemiphyllum var. chinense were 0.65mg/g, 0.46mg/g, 1.01mg/g, all of which had branched chain amino acids.In terms of cell survival, the toxicity of brown algae extraction to mouse skeletal muscle fibroblasts (C2C12): S. horneri  (5mg/mL) , S. hemiphyllum var. chinense (5mg/mL) , S. ilicifolium (10mg/mL).When the dose concentration is 5mg/ml atS. horneri , S. hemiphyllum var. chinense and S. ilicifolium , the cell survival rate was 82.24%, 97.22% and 136.6%. In the mTOR test, the concentration of three kinds of brown algae (S. horneri , S. hemiphyllum var. chinense and S. ilicifolium ) were 97.2pg/mL, 21.72pg/mL and 100.52pg/mL to increase compared to non-added groups, and each pointer has dosedependent.
