台灣重要河川魚介類遺傳資源分析與評估

AFLP分析技術於1995年研發後,已成為近年來研究趨勢;本研究擬利用HindⅢ及EcoRⅠ限制酶在核酸上不規則分佈,並以特定接引子(Adapter)處理後,利用緊臨限制酶切點的2個任意核苷酸作為篩選,以225組逢機組合進行PCR反應,擴增DNA片段於電泳膠上呈現一特定的基因圖譜,此技術平台可作為種內與種間之分析研究,亦可作為種源庫資訊與參考。

AFLP technique, developed by Vos et al. (1995), is based on the selective PCR amplifiedof restriction fragments from a total of genomic DNA. To research and identifynumbers of AFLP markers, 225 primers combinations, base on the restriction enzymeEcoRI and HindⅢ with two base pair extensions, were tested to generate AFLP pattern.The markers are suitable for use in gene pool and trait loci by using the inter- and intraspecies.Analysis of genetic variations among strains of fish indicated that AFLP shouldalso be a valuable tool for genetic resource and population analysis in fish.
