The nervous necrosis virus (NNV) and iridovirus (IRV) have been attributed to a measurable mass mortality of the grouper. This study aims to develop the techniques to improve the effect of vaccine to induce protection of grouper against virus infection. The results showed that the safety injection IRV vaccine was 0.01 to 0.2 ml/fish to the grouper fry (4-6 cm). In the challenge test, the survival rate of the vaccine groups of fish challenged with injection of IRV diluted solution was higher (p<0.01) than control group fish over 50% after IRV vaccine treatments for 2, 4 and 6 weeks. In the neutralization test of test fish serum collected in different period after injection of IRV vaccine showed that the highest title of grouper serum was appeared at 2 weeks after vaccination and last over 8 weeks.
