The specific pathogen free (SPF) broodstock of Penaeus monodon with White Spot Syndrome Virus (WSSV)、Taura syndrome virus(TSV) and Infectious Hypodermal and Hematopoietic Necrosis Virus (IHHNV) were sieved from wild species for breeding SPF larvae. The six sexual mature female P. monodon were wild-caught broodstock of Tung Kang, and four of the six were SPF following PCR examination. Moreover, after sieving and PCR examination, 50 pairs of adult P. monodon with ablation eyestalk were conducted from the cultivating ponds. The amount of 200,000 larvae from the four wild species P. monodon were sieved and examined for breeding, and the same procedure was carried in the cultivated ones. Following the developing stages, the larvae were sieved during cultivation.
The larvae were challenged for sieving high tolerance to virus infection for further cultivating. The two resources of larvae were randomly sampled for challenging tests according to each individual brood stock. The challenging test consisted of WSSV infection and tolerance to hypothermal exposure. The highest survival rate of WSSV challenging test was 36.3%, and the lowest one was 18.3%; the highest survival rate under hypothermal exposure was 21.6%, and the lowest one was 8.1%.
