海洋機能素材之研發及其功效性探討(3)龍鬚菜類蕈孢 素胺基酸之安全性及功能性評估

類蕈孢素胺基酸(Mycosporine-like amino acids, MAAs)是海洋生物體內具有吸
收UV光的機能性成分,在本計畫中我們將萃取自龍鬚菜的類蕈孢素胺基酸進行安全
性及功能性評估,希望在瞭解其安全劑量後,再開發其應用性,以增加龍鬚菜的多
元化利用。本研究使用安姆測試 (Ames Test) 進行安全性評估,其中選用5株
Samonella typhimurium做為測試菌株。另以抗光老化細胞試驗進行功能性評估,其
中選用皮膚纖維母細胞進行UVB光損傷,進而分析類蕈孢素胺基酸對細胞存活率、胞
內自由基及基質金屬蛋白酶(Matrix Metalloproteinases, MMPs)表現量。最後
,建立單次進料20 kg龍鬚菜(乾重)之前處理、萃取條件及相關機械設備之總耗時
、收率…等量化條件。

Mycosporine-like amino acids (MAAs) are a family of intracellular
UVabsorbing compounds involved in the protection of marine organisms
against solar UV radiation. In this study, we focus on the safety and
functional evaluation of MAAs extracted from Gracilaria. The safety
evaluations of MAAs were determined in the Ames test, which was conducted
by the plate incorporation method using five strains of Samonella
typhimurium. The functional evaluations of MAAs were determined in the
anti-aging actions of MAAs on UV-induced aging in skin fibroblasts. The
fibroblasts were pretreated with MAAs. After UVB irradiation, the levels
of aging were determined by measuring MTT assay, Intracellular ROS
content and the expression of matrix metalloproteinases. Finally, we
estimate the condition of preparation, extract process and equipments for
20 kg Gracilaria (dry base) per batch.
